Test principle : This assay employs a two-site sandwich ELISA to quantitate TMEM171 in samples. An antibody specific for TMEM171 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTMEM171 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for TMEM171 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TMEM171 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : TMEM171 (transmembrane protein 171) is a 324 amino acid protein encoded by a gene mapping to human chromosome 5. With 181 million base pairs encoding around 1000 genes, chromosome 5 is about 6% of human genomic DNA. It is associated with Cockayne syndrome thro µgh the ERCC8 gene and familial adenomatous polyposis thro µgh the adenomatous polyposis coli (APC) tumor suppressor gene. Treacher Collins syndrome is also chromosome 5 associated and is caused by insertions or deletions within the TCOF1 gene. Deletion of the p arm of chromosome 5 leads to Cri du chat syndrome. Deletion of 5q or chromosome 5 altogether is common in therapy-related acute myelogenous leukemias and myelodysplastic syndrome.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
